Tissue Procurement: Human intervertebral disc (IVD) tissue is collected from the spine of cadaveric donors within 24 hours of death. Proper consent and serological information is obtained on all specimens. The intervertebral disc tissue is harvested into two pools, the nucleus pulposus (NP) and annulus fibrosus (AF); intermediate tissues are discarded.
Alginate Bead Processing: Disc tissue is digested with a sequence of enzymes to remove disc cells from surrounding matrices. The cells are combined with alginate gel and added drop-wise into a calcium salt bath to initiate polymerization. Alginate beads are recovered and distributed into multi-well plates or into the desired format with 50/50 DMEM/F12 media supplemented with 10% FBS. Alginate bead cultures are shipped with ice packs for overnight delivery. |
