Tissue Procurement: Human synovial tissue is collected from cadaveric donors within 24 hours of death or as surgical specimens from a relevant procedure. The tissue is harvested from the knee joint from both clinically “normal” donors and from those diagnosed with rheumatoid and osteoarthritis. Proper consent and serological information is obtained on all specimens.
Explant Processing: Synovial tissue is carefully excised from surrounding fat and tissues. Explants are cut to approximately 3mm in size and weigh between 10 and 20mg. Due to the nature of synovial tissue all sizes are estimated and it is recommended that wet weights be determined for normalization purposes. Explants are distributed into multi-well plates or in bulk format in 50/50 DMEM/F12 media supplemented with 10% FBS. The cultures are shipped overnight with cold packs. |
