Tissue Procurement: Human intervertebral disc (IVD) tissue is collected from the spine of cadaveric donors within 24 hours of death. Proper consent and serological information is obtained on all specimens. The intervertebral disc tissue is harvested into two pools, the nucleus pulposus (NP) and annulus fibrosus (AF); intermediate tissues are discarded.
Monolayer Culture Processing: Disc tissues are treated with a sequential enzyme cocktail to isolate cells from surrounding matrices. The cells are counted and transferred to a flask or multi-well plate in 50/50 DMEM/F12 media supplemented with 10% FBS. The cells are maintained in a humidified incubator at 37oC and 5% CO2 for 48 hours to promote cell adhesion and then shipped with cold packs for overnight delivery. |
