Tissue Procurement: Bovine bone tissue is collected from the end of long bones surrounding the metacarpalphalangeal joint (“knuckle”) of 18 month old cows.

 

Monolayer Culture Processing: Harvested bone tissue is carefully dissected away from surrounding tissues and washed thoroughly to remove blood and debris. An enzyme treatment is applied to isolate primary bone cells. The cells are seeded onto flasks or multi-well plates in Ham’s F12 media supplemented with 10% FBS. The cells are maintained in a humidified incubator at 37^oC and 5% CO₂ for 48 hours to promote cell adhesion and then shipped with cold packs for overnight delivery.
